It means the set "snakes" is contained in a 2-dimensional space...literally, "Snakes on a Plane."
I actually got it as soon as I saw it.
But I didn't want to be the smart-aleck geek kid sitting in the front row and raising his hand and blurting out the answer before everyone else even had a chance to finish reading it.
My wife and I have been to at least two different movies now where they showed the preview for "Snakes on a Plane", and both times I think we gave each other the same puzzled look, like "What the...? They made a whole movie out of that?"
(Of course now I get to be the guy who shows up after the answer was revealed and says "Hey, I knew that!")
